Vitamin D2 vs D3: What's the Difference?

    It is one of the most-Googled questions in supplement-shopping: vitamin D2 versus vitamin D3, what is the difference, and does it actually matter. The short answer is that it matters, the research consensus is reasonably clear, and most modern supplements have settled on D3 for sensible reasons. The longer answer covers why — and what to do if you specifically need a vegan source.

    The two forms

    Vitamin D comes in two principal forms, both produced in nature by the action of ultraviolet light on a precursor molecule.

    Vitamin D2 (ergocalciferol) is the plant-derived form. It is produced by certain fungi, lichens and yeasts when exposed to UV light. Historically, the cheap industrial source of D2 was irradiated ergosterol from yeast or fungi. Most prescription high-dose vitamin D in the UK and US has, until recently, been D2 because it was cheaper to produce at scale.

    Vitamin D3 (cholecalciferol) is the animal-derived form — the same molecule your skin produces when exposed to UVB radiation, and the form found in oily fish, egg yolks, liver and (in smaller amounts) red meat. Most commercial D3 comes from sheep's lanolin, although vegan D3 derived from lichen is increasingly available.

    Both forms, once absorbed, undergo a two-stage conversion in the body — first in the liver to 25-hydroxyvitamin D (the storage form measured in blood tests), then in the kidneys (and locally in many tissues) to 1,25-dihydroxyvitamin D, the active hormone.

    Are they equally effective?

    This is where the research has moved. For a long time, the assumption was that D2 and D3 were biologically equivalent — both raise vitamin D levels, both function as vitamin D in the body. More recent and better-designed research has found this is not quite true.

    A widely cited 2012 meta-analysis by Tripkovic and colleagues in the American Journal of Clinical Nutrition pooled the available randomised controlled trials comparing D2 and D3 supplementation. The conclusion: D3 was significantly more effective at raising and maintaining serum 25-hydroxyvitamin D levels than D2, particularly when given as a single large dose.

    The mechanism appears to be twofold. D3 binds more tightly to vitamin D binding protein, which transports it through the bloodstream, meaning it stays in circulation longer. D3 also appears to be converted to 25-hydroxyvitamin D more efficiently by the liver. The result is that D3 raises blood levels more, and keeps them raised for longer, than equivalent doses of D2.

    The UK's SACN 2016 report reviewed the literature in detail and concluded that, while both forms count toward dietary intake, D3 appears to be more potent.

    How big is the difference, practically?

    The Tripkovic meta-analysis found D3 to be roughly twice as effective as D2 at raising serum 25-hydroxyvitamin D, particularly with bolus dosing. With smaller daily doses, the gap is narrower but still present. The practical implication is that if you take 1,000 IU of D2 and 1,000 IU of D3, the D3 will, on average, lift your blood levels more.

    There is also a stability angle. D3 appears to be more stable in supplement form than D2, particularly in formulations exposed to heat or humidity. This is a smaller issue with well-manufactured products but matters at the margins.

    What the NHS recommends

    The NHS recommends a daily 10 microgram (400 IU) vitamin D supplement for everyone over the age of one through autumn and winter months. The NHS does not specify D2 or D3, but in practice the vast majority of pharmacy and supermarket vitamin D supplements in the UK are now D3, and this is the form most clinical research supports.

    The SACN's view, summarised in the 2016 report: "It is recommended that vitamin D3 is used in preference to vitamin D2 when supplementing."

    What about vegan considerations?

    This is the one significant practical reason someone might choose D2 over D3. Historically, the only animal-free form was D2, which made it the default choice for vegans, vegetarians who avoid animal-derived supplements, and certain religious dietary practices.

    This is no longer the case. Vegan D3 derived from lichen is now widely available and produces the same form of the molecule as conventional lanolin-derived D3 (or skin-synthesised D3). It is more expensive to produce but bioequivalent to standard D3. If you are vegan or avoid animal-derived supplements, vegan D3 is a better choice than D2 from an effectiveness standpoint.

    What about safety?

    Both forms are safe in normal supplemental doses. The European Food Safety Authority has set the safe upper intake level for vitamin D in adults at 100 micrograms (4,000 IU) per day. Standard supplements deliver well below this. Toxicity from vitamin D supplementation is rare and usually requires very large doses taken consistently over time.

    Anyone with kidney problems, certain medical conditions, or taking medication that affects calcium metabolism should speak to their GP before starting high-dose supplementation. For most adults taking the NHS-recommended 10 micrograms daily, there is no realistic safety concern.

    What about absorption and timing?

    Both D2 and D3 are fat-soluble. They are absorbed more efficiently when taken with food containing some fat. This is partly why supplements are often recommended at breakfast, when most people are eating something with at least small amounts of fat (butter on toast, olive oil, milk in coffee, etc.). Taking a fat-soluble vitamin D supplement on an empty stomach is not optimal, regardless of form.

    There is no strong evidence that morning vs evening timing matters significantly for vitamin D supplementation, although some people prefer morning dosing for consistency. The most important thing is that you actually take it — building a habit matters far more than timing optimisation.

    The vitamin D system is more than just D

    One thing worth noting in the D2-vs-D3 discussion: focusing exclusively on the form of vitamin D misses that vitamin D in the body operates as part of a system. Once D3 is converted to active calcitriol, its effects depend on:

    • Vitamin K2, which directs the calcium vitamin D absorbs toward bones rather than soft tissues
    • Magnesium, which is required as a cofactor for vitamin D activation
    • B vitamins, zinc and selenium, which participate in the parallel systems vitamin D regulates

    A high-quality supplement, especially for British indoor lives, considers the whole system. D3 alone is better than D2 alone. D3 with K2, magnesium and the supporting cast is better than D3 alone.

    The bottom line

    If you are choosing between D2 and D3, choose D3. The research is clear that D3 raises blood vitamin D levels more effectively, the SACN explicitly recommends it, and the vegan-friendly version is widely available.
